DescriptionSiglec-15, a member of the Siglec family of sialic acid-binding Ig-like lectins, is a type I membrane protein primarily expressed in M2-type macrophages, myeloid-derived suppressor cells (MDSCs), dendritic cells, B cells, and osteoclasts. Studies have shown that for most PD-L1 negative tumor patients, siglec-15 is likely to be the main immune inhibitory factor. This protein is mutually exclusive with PD-L1, meaning that the expression of PD-L1 and Siglec-15 in tumor cells decreases as one increases. Siglec-15 can continuously inhibit T cell activity and exhibits the main characteristics of normalizing cancer immunotherapy. For those patients with psoriasis who are resistant to PD-1/PDL1 therapy, antibodies that block the interaction between Siglec-15 and CD33L3 may have immunotherapeutic effects.
