Description: TIGIT (T cell immunoglobulin and ITIM domain), also known as WUCAM, Vstm3, VSIG9, is a member of the PVR/Nectin family. It consists of an extracellular immunoglobulin variable domain (lgV) structure domain, a 1-type transmembrane domain, and an intracellular domain with classical immunoreceptor tyrosine inhibitory motifs (ITIMs) and immunoglobulin tyrosine tails (ITTs). TIGIT is expressed in lymphocytes, especially in effector and regulatory CD4+T cells, follicular helper CD4+T cells, effector CD8+T cells, and natural killer (NK) cells. So far, it has been found that TIGIT can interact with CD155, CD112, and CD113, among which the interaction between TIGIT-CD155 is the strongest. CD155 is mainly expressed on the surface of immune cells such as DC cells, T cells, B cells, macrophages, etc. In non-immune cells such as kidneys, lungs, and pancreases, there are also a small number of expressions. CD112 is widely expressed on the surface of hematopoietic system cells and non-hematopoietic system cells, but CD113 is only expressed on the surface of non-hematopoietic system cells. What's worth noting is that CD155 and CD112 are highly expressed on the surface of many malignant tumors such as colorectal cancer and melanoma.
