Apolipoprotein E (ApoE) is a polymorphic protein that serves as one of the most crucial lipid transport proteins in the human body. It is also the apolipoprotein with the greatest impact on lipid metabolism, primarily involving triglycerides and cholesterol. ApoE is distributed throughout most tissues in the human body and is mainly found in chylomicrons (CM), very low-density lipoproteins (VLDL), intermediate-density lipoproteins (IDL), and some high-density lipoproteins (HDL).
Apolipoprotein E3 (ApoE3) is the most common isoform of ApoE, characterized by cysteine-arginine residues at positions 112 and 158. It plays a central role in lipid metabolism by mediating the binding of lipoproteins (e.g., VLDL, HDL) to LDL receptors and heparan sulfate proteoglycans. ApoE3 is critical for cholesterol transport and neuronal repair in the central nervous system.
