Ompekimig is a immunoglobulin trispecific antibody that targets IL-4, IL-13 and IL-33.
A key Th2 cytokine produced by T cells and mast cells, IL-4 drives B cell class-switching to IgE, promotes M2 macrophage polarization, and mediates anti-parasitic and allergic responses. It signals through the IL-4Rα/γc receptor complex. Closely related to IL-4, IL-13 shares the IL-4Rα receptor subunit and contributes to allergic inflammation, airway hyperresponsiveness, and tissue fibrosis. It directly acts on epithelial cells and smooth muscle, enhancing mucus production and remodeling. An alarmin from the IL-1 family, IL-33 is released by damaged epithelial/endothelial cells. It activates ST2 receptor-expressing immune cells (e.g., ILC2s, Th2 cells), amplifying type 2 immunity and driving allergic and fibrotic pathologies.
