Lofacimig is an only-heavy-chain immunoglobulin bispecific antibody that targets F11 apple 2 domain and F11 apple 3 domain. Factor XI (FXI) is a clotting protein with four tandem Apple (A) domains (A1-A4). These domains mediate key interactions for FXI activation and function. The A2 domain docks FXI to platelets via HK/heparin, while the A3 domain captures and activates FIX. Together, they ensure targeted, efficient clot formation.
